Hi, I will complete all my degree requirements before April 1st, but I will graduate only in May. Can I still file H1B Petition in the new quota on April 1st?

This should not be a problem as the requirement is you should be eligible. If you have completed the degree requirements, you are qualified for an H1B job.

I know someone in this situation who got H1B visa last year.

You will need to get the necessary documentation from the appropriate authority at your University attesting that you have completed all the degree requirements and submit that along with the H1B Petition. We have had several H1B Petitions approved with such documentation.
